Interferon gamma is a multifunctional protein produced by cells of the immune system (T – cells and NK – cells), has immunostimulating (activates T-cells), antitumor (antiproliferative) effect– by increasing the number of molecules of MHC I and induction of antiangiogenic effect by secretion of interferon-dependent-gamma IP — 10 and MIG – and antiviral activity-activates macrophages by increasing the number of MHC II molecules.
Creation of microorganisms-producers of human interferon gamma allowed to receive biologically active recombinant protein in the quantity necessary both for carrying out wide structural and functional researches, and for use in medical practice.
A strain of Escherichia coli — a producer of human interferon gamma and a method for obtaining a highly purified preparation of unprocessed non-methionine native recombinant human interferon gamma have been developed.
The resulting highly purified preparation of native human interferon gamma has a high specific biological activity (2 000 000 units per 50 µg) and stability. At least 95% of the basic substance of the drug is in the form of a monomer, and in the dimer form — no more than 5%. The resulting drug reacts only with antibodies to interferon gamma (Peprotech company, catalog number 500-M90), stable in the pH range from 4.5 to 10. Specific antiviral activity is not less than 2*106 ME / mg.
